ASO-Turbilatex
DIAGNOSTIC SIGNIFICANCE:
SLO is a toxic immunogenic exoenzyme produced by (3-hemolytic Streptococci of groups A, C and G. Measuring the ASO antibodies are useful for the diagnostic of rheumatoid fever, acute glomerulonephritis and streptococcal infections. Rheumatic fever is an inflammatory disease affecting connective tissue from several parts of human body as skin, heart, joints etc… and acute glomerulonephritis is a renal infection that affects mainly to renal glommerulus.

PRINCIPLE:
The ASO-Turbilatex is a quantitative turbidimetric test for the measurement of ASO in human serum or plasma.Latex particles coated with streptolysin O (SLO) are agglutinated when mixed with samples containing ASO & the agglutination causes an absorbance change, dependent upon the ASO contents of the patient’s sample that can be quantified by comparison from a calibrator of known ASO concentration.

CLINICAL SIGNIFICANCE:
There are two forms of cholinesterase; acetyl cholinesterase and cholinesterase or also commonly referred to as psuedocholinesterase. Acetylcholinesterase is found predominantly in erythrocytes. Cholinesterase is synthesized in the liver and is present in plasma and is the form of the enzyme routinely measured. Cholinesterase is most commonly measured as an indicator of exposure to anticholinesterases (organophosphates, including many insecticides), or inherited abnormal variants of the enzyme, which cause a decreased level of plasma cholinesterase.Increased levels of activity may be present in nephrotic syndrome or in the recovery from liver damage.

PRINCIPLE:
Cholinesterase hydrolyses propionylthiocholine to propionic acid and thiocholine. Thiocholine reacts with 5, 5′-dithio-bis (2-nitrobenzoic acid) to form the yellow coloured 5-thio -2-nitrobenzoic acid. The rate of formation of 5-thio-2-nitrobenzoic acid, measured at 405nm, is directly proportionalto cholinesterase activity in the sample. BACKGROUND AND SYNOPSIS:
Conventional methods like Ferricyanide, Nelson-Somogyii, Folin-Wu, O-Toludine involved lengthy procedures and were cumbersome. Some of these methods involved steps like precipitation, filteration and boiling. These methods have drawbacks, are non-specific, imprecise, insensitive and inaccurate.ENZOPAK Glucose is based on GOD/POD method (Glucose oxidase /Peroxidase) as described by Trinder and is specific and accurate. The method selectively estimates β-D-Glucose in serum or plasma.

PRINCIPLE:
Glucose oxidase oxidises the specific substrate, β-D-glucose, to gluconic acid and hydrogen peroxide is generated. Hydrogen-peroxide thus produced is acted upon by peroxidase and oxygen is liberated. The liberated oxygen is transferred to chromogen system consisting of 4-aminoantipyrine and phenolic compound to produce red quinoneimine dye. The intensity of colour is directly proportional to the concentration of glucose and is measured photometrically at 505 nm(500-540 nm or with GREEN filter)
