2-GLYCOPROTEIN I IGG, ELISA
The DIAWell β2–Glycoprotein I IgG kit is a solid phase enzyme immunoassay using 96 coated breakaway microwells and a peroxidase-TMB detection system. The microwells are coated with highly specific antigen. In the test procedure, serum samples are diluted 1/51 and incubated in the microwells. Human antibodies, if present, bind to the specific antigen. Unbound or excess antibodies are removed by washing and HRP-conjugated rabbit antibodies against human IgG are added to the microwells. The enzyme conjugate binds to the antigen-antibody complexes. After a second washing step to remove excess conjugate, the TMB/substrate solution is added. The enzyme activity, if present, generates a colorimetric (blue) reaction. Diluted acid is added to stop the reaction. Consequently the colour turns from blue to yellow and may be measured at 450 nm using a conventional microplate reader. The absorbance (Optical Density) is directly proportional to the concentration of IgG antibodies bound to the antigen on the microwells surface

SEROTONIN, FASTRACK ELISA
In the first step, Serotonin is quantitatively acylated. The subsequent competitive ELISA kit uses the microtiter plate format. The antigen is bound to the solid phase of the microtiter plate. The acylated standards, controls and samples and the solid phase bound analyte compete for a fixed number of antiserum binding sites. After the system is in equilibrium, free antigen and free antigen-antiserum complexes are removed by washing. The antibody bound to the solid phase is detected by an anti-rabbit IgG-peroxidase conjugate using TMB as a substrate. The reaction is monitored at 450 nm. Quantification of unknown samples is achieved by comparing their absorbance with a reference curve prepared with known standard concentrations.

SEROTONIN, RIA
First, Serotonin is quantitatively acylated. The assay procedure follows the basic principle of radioimmunoassay, involving competition between a radioactive and a non-radioactive antigen for a fixed number of antibody binding sites. The amount of 125I-labelled antigen bound to the antibody is inversely proportional to the analyte concentration of the sample. When the system is in equilibrium, the antibody bound radioactivity is precipitated with a second antibody in the presence of polyethylene glycol. The precipitate is counted in a gamma counter. Quantification of unknown samples is achieved by comparing their activity with a reference curve prepared with known standards.

MUMPS IGG, ELISA
The KAPRMUG12 Mumps IgG kit is based on the ELISA technique. In the assay, controls and unknowns are incubated in microtitration wells coated with purified and inactivated Mumps Virus antigen. After incubation and washing, the wells are treated with the conjugate, composed of anti-human IgG antibodies labeled with peroxidase. After a second incubation and washing step, the wells are incubated with the substrate tetramethylbenzidine (TMB). An acidic stopping solution is then added and the degree of enzymatic turnover of the substrate is determined by wavelength absorbance measurement at 450 nm. The absorbance measured is directly proportional to the concentration of anti-Mumps virus IgG antibodies present.

ZIKA VIRUS IGM Μ-CAPTURE, ELISA
The qualitative immunoenzymatic determination of specific IgM-class antibodies is based on the ELISA (Enzyme-linked Immunosorbent Assay) μ-capture technique. Microplates are coated with anti-human IgM-class antibodies to bind the corresponding antibodies of the sample. After washing the wells to remove all unbound sample material a horseradish peroxidase (HRP) labelled Zika virus antigen is added. The immune complex formed by the bound conjugate is visualized by adding Tetramethylbenzidine (TMB) substrate which gives a blue reaction product. The intensity of this product is proportional to the amount of specific IgM antibodies in the sample.
